Hell Fucking Yes.

I second that Hell Fucking Yes. Had the good fortune of seeing her play with the Bush Tetras once, and she rained genius for an hour. She's nothing short of heroic on a good night.

And big up to Brett Ralph for namechecking the equally righteous Thalia Zedek.

And while we're citing cool women, Poison Ivy fron the Cramps always rocks the house live -- one of my fave players in the rockabilly pantheon. And the late Larissa Strickland from the Laughing Hyenas cranked out some of the most fearsome guitar I have ever heard in my life. Her work on Life Of Crime will rattle your windows at a decent volume.

My first pick is Neil Young, of course, subtle as fuck rhythm player, and the king of the one note solo....

I would go see a band called Subtle As Fuck.
I knew I'd be way behind on mentioning Lindsey Buckingham.
I was going to say James Honeyman-Scott but then I decided he wasn't really underrated, just a guitarist I liked.
If the Stone Roses' plane had gone down after their first LP I would definitely say John Squire. As it is, I still might.
I haven't heard enough '80s cock-rock heroes in this thread, so: is Steve Stevens underrated? Just putting it out there.
The "can girls play guitar?" sub-thread got me thinking: maybe Nancy Wilson? (http://youtube.com/watch?v=AcAn3z7D1Pc)
... and definitely Dolly Parton. "And people say she's just a pair of tits."

Mary Timony thinks you're a jackass.

that woman is an absolute wizard on guitar. the whole chord/two note finger pick/ ending in a bent harmonic and moved up and down the neck thing is so consistently mindblowing. I'd give up one of your legs to play like her.
kerble is right.
